I would say make a bracket like you would for a hard saddle bag. Get a hard shell case to mount to the bracket. Put your guitar in a soft case that will insert into the hard case.
I have a padded soft case that I used to wear on my back when I cycled to some local gigs.Should be fine on a motorcycle up to say 50/60 Mph.

And even beyond that if you are careful. I drove 173 miles to my cabin in Ottertail, MN with my acoustic strapped to my back in a soft case. I drove 70-75 on the interstate. I was a little concerned, but the bag held up. Granted, I was rolled up into the fetal position with my clip-on's, so I am not sure how you would fair sitting up. After 3 hours my back was killing me...
